As far as we know, Jared Padalecki is not involved in The Winchesters at all. Jensen Ackles is heavily involved in the series, but Padalecki is extremely busy starring in the CW's Walker and he is also working on a prequel to that show. Chances are, Padalecki will not be reprising his role as Sam Winchester in The Winchesters, but who knows?
Misha Collins Isn't In It
Misha Collins's Castiel is undoubtedly one of the most beloved Supernatural characters. Making his debut in Season 5, Castiel serves as Sam and Dean's guardian angel, partner, accomplice, and friend. In fact, fans have shipped Castiel and Dean for a long time.

Since Supernatural premiered in 2005, a lot of diehard fans are much older than the intended target audience for The Winchesters. The CW is a bonafide teen network, and the only reason why Supernatural escaped that reputation is that it was originally a Warner Brothers production.
The Winchesters already looks like a Riverdale-esque teen drama, and that's unappealing to a lot of Supernatural fans who don't necessarily want to watch an overly dramatic, mushy show about two teenagers in love. The main purpose of the show is to draw in new viewers, not necessarily to please old fans.
Fans Would Rather Hear Other Supernatural Stories
Nobody ever asked for a show about John and Mary Winchester. These characters are not widely loved by Supernatural fans, as they honestly aren't great parents. There are so many other characters that fans would much rather get a show about, but it doesn't seem like that's going to happen anytime soon.
Supernatural has introduced a lot of interesting, complex, and beautiful characters who could star in their own spinoff shows. In fact, Sheriff Jody Mills and all her adopted daughters were initially supposed to receive their own show, Wayward Sisters, but it ended up getting canceled because the CW thought people wouldn't watch it. There are a lot of other characters that fans would love to learn more about, but John and Mary aren't necessarily on that list.
