While the new trailer for Spider-Man: No Way Home has confirmed five different multiversal villains, who could the sixth Sinister Six member be? No Way Home's second trailer has been released, revealing that a ruined spell cast by Doctor Strange will bring villains from across the multiverse to the MCU. However, it seems as though one final member is missing from the line-up of what would be a multiversal Sinister Six Peter Parker will need to face.

In the second trailer for Spider-Man: No Way Home, Benedict Cumberbatch's Doctor Strange confirms that the spell he cast (which Peter accidentally ruined) has brought "visitors from every universe." The already confirmed Doctor Octopus played by Alfred Molina is seen once more, discovering that Tom Holland's Spider-Man is not the Spider-Man he knows (Tobey Maguire). Furthermore, the Sorcerer Supreme reveals that he and Spider-Man need to round up these villains and send them back, confirming that they're all foes who were killed while fighting different versions of Spider-Man in their own universes. With that, Jamie Foxx's Electro is seen in the trailer along with the confirmation of Willem Dafoe's Green Goblin, as is the version of the Lizard seen in The Amazing Spider-Man starring Andrew Garfield, while Sandman brings the total number of villains to five.

However, Spider-Man's greatest challenges in the comics have always come from the Sinister Six, so who might a sixth and final villain be? Given Doctor Strange's reveal in the trailer that all of the villains featured thus far are foes who are killed by a Spider-Man in their respective universes, perhaps the final villain could be Jake Gyllenhaal's Mysterio. Mysterio already will be important in No Way Home given the public's belief that he died a hero before outing Spider-Man as Peter Parker and a menace, and he did seemingly die from one of his drone's gunfire while trying to kill Peter in Far From Home. Mysterio was the reason Peter even approached Strange in the first place, wanting the Sorcerer Supreme to undo Quentin Beck revealing his identity to the world. As a result, it would be fitting to have him be the final member of this new Sinister Six coming to the MCU.

However, an alternate option could be Tom Hardy's Venom. The post-credits scene in Venom: Let There Be Carnage featured the Lethal Protector seemingly falling victim to a shift in the multiverse where he's now in the same universe as Tom Holland's Spider-Man, seemingly recognizing the Webslinger via the symbiote's connection to a hivemind which spans universes. While Tom Hardy's Venom has never met Spider-Man (much less been killed while fighting him), Topher Grace's Venom from Tobey Maguire's Spider-Man universe did, so the current Venom could technically fit the criteria as the other villains seen in No Way Home's new trailer.

In any case, it's going to be interesting to see how Tom Holland's Spider-Man copes with all of theses villains who will apparently want to stay in the MCU, rather than meeting their demises back in their own universes. Furthermore, Peter Parker is just one Spider-Man facing five very deadly villains who will most likely be joined by a just as powerful sixth. Regardless of whether it's Mysterio, Venom, or some other foe entirely, Spider-Man: No Way Home is being set up to be one of the MCU's biggest confrontations.
